Verification of the Cary 60 UV-Vis spectrophotometer for quantification of bilirubin and hemoglobin derivatives in cerebrospinal fluid (English)
Abstract [en]

The purpose of the study was to verify the spectrophotometer Cary 60 UV-Vis from Agilent for the analysis of bilirubin and hemoglobinderivatives in cerebrospinal fluid. The values were compared with those obtained using the current spectrophotometer Genesys 10S from Thermo Scientific.

A total of 31 patient samples with low and high concentrations of bilirubin and hemoglobinderivatives were analyzed. A baseline stability test was performed in which deionized water was analyzed 10 times. Precision was evaluated by analyzing one CSF sample 10 times. Eight filters were used to analyze wavelength and absorbance accuracy. Spectral scanning was carried out within the wavelength range of 350–600 nm.

The mean value and standard deviation for baseline stability for both spectrophotometers were close to 0. For the precision analysis, a CV% of 0.06% was calculated. All r-values were above 0.9. R² values were above 0.9 except for samples with low concentrations. Bias for all samples was close to zero, with limits between 0.162 and −0.126. Sources of error such as the limited number of samples may have affected the results.

Based on the results it can be concluded that the new spectrophotometer can replace the current spectrophotometer.
